Protein Expression and Characterization

The Protein Expression and Characterization facility offers scientific expertise and technical assistance to help researchers in all aspects of the production of recombinant proteins. The workflow for typical protein expression project includes: construct design and cloning, small-scale expression tests, scale-up protein production, purification and quality assessment. The facility provides a complete service, from construct design to purified product. This approach allows selecting a strategy, identifying optimal expression conditions and optimizing purification protocol. Target proteins can be expressed in prokaryotic and eukaryotic hosts such as Escherichia coli or baculovirus/insect cells expression systems (use of mammalian cells is considered in future). The facility is using a versatile set of vectors enabling multi-host expression without need for subcloning. A range of fusion partners, which may be removed after initial purification, is available to improve solubility of the over-expressed target protein. Facility supported biochemical and biophysical characterization includes dynamic light scattering, size exclusion chromatography with multi-angle light scattering, differential scanning fluorimetry and analytical ultracentrifugation.
